Abstract
:
Sidr is one of the most common medicinal plants frequently used in folk medicine , especially in Saudi Arabia . This study aims basically to identify influence of daily treatment by Sidr Leaves and Fruits Extract in total content of neurotransmitters (norepinephrine , dopamine, serotonin and gammaaminobutyric acid ) in different brain regions : (Cerebellum - brainstem - cerebral cortex – hypothalamus and hippocampus ) in male albino rat , in addition to know the role of possible protective and therapeutic of Sidr leaves and fruits extract against convulsions caused by substance pentylenetetrazol. Chronic injection by leaves extract (50 mg/kg i.p.) and a gradual withdrawal increasing neurotransmitters content in most different brain regions at most of time interval , this is probably due to the presence of cyclic peptides and peptides alkaloids in the extract which worked on the inhibition of Ca2+/ calmodulin protein binding which plays an important role in release of these neurotransmitters , on the other hand saponin discourage monoamines release from secretary of cells that produce them , so the content is increased. The study also showed that Chronic injection by fruits extract (50 mg/kg i.p.) and a gradual withdrawal decreasing neurotransmitters content in most different brain regions at most of time interval , which could be due to the presence of ascorbic acid which caused neurotransmitter release from presynaptic by enhanced Ca2+ influx , also ascorbic acid increase nitric oxide (NO) levels , so the content of neurotransmitter is decreased . The results also showed a clear improvement in the total content of neurotransmitters in treated animals by protective and therapeutic dose of Sidr leaves and fruits extract which decreased significantly by treatment with pentylenetetrazol , this is probably due to sidr leaves are rich material saponin that worked to inhibit the release neurotransmitters of cells that produce them which has been activated with pentylenetetrazol , also , the sidr fruits rich in ascorbic acid, which work to discourage the activity of the Na, K-ATPase leading to inhibition of nerve conduction and then inhibition of neurotransmitter release and increased content in the brain. The study concludes that leaves and fruits extract may have protective and therapeutic role as anticonvulsant effect , this needs to be further research and study .
